研究内容

2.表观遗传。有些环境刺激(如饮食、毒素、应激、情绪)虽不诱发基因突变,却可持久地改变基因功能。这些改变有时可以遗传给后代,以致决定我们表型的,不但有基因,还有祖辈的生活经历甚至喜怒哀乐。这种惊人的遗传现象颠覆着经典遗传学,但其物质基础扑朔迷离。我们建立了最简易的小鼠模型,来攻克这一难题。

2.遗传学技术。工欲善其事,必先利其器。我们因此研发了多种小鼠遗传学工具,聚焦高通量筛选。对大多数基因,尤其是近年发现的LncRNA,我们只知序列不知功能,“知人知面不知心”。高通量筛选是揭开基因真面目的强大手段,但筛选只能在体外进行,这就严重地限制了其用途并构成了遗传学一重大瓶颈,因为多数生理现象无法在体外复制。我们试图用一种崭新的方法(Cre + Cas 9) 来打破这一瓶颈, 并用它揭示基因尤其是LncRNA 在小鼠体内的功能。
